PEPTIDE BASED IMMUNODIAGNOSTIC APPROACH IN THE DIAGNOSIS OF
NEUROLOGICAL DISEASE.

Clinical immunology is the study of diseases caused by disorders of the immune system. The specificity of the bond between antibody and antigen has made the antibody an excellent tool for the detection of substances by a variety of diagnostic techniques.

In CIIMS, we have developed several immunological assays for the detection of antigens and antibodies against various bacterial diseases like Pulmonary and Extra Pulmonary TB; viral diseases like Chikungunya, Herpes Simplex virus Encephalitis, Cytomegalovirus Encephalitis, Epstein Barr virus Encephalitis, Varicella Zoster virus Encephalitis, Japanese virus encephalitis, Dengue virus, Chandipura virus and West Nile virus Encephalitis and Fungal diseases like Cryptococcal Antigen. Also developed a screening test for diagnosis of deadly infection i.e. Scrub Typhus. These all above mentioned tests are developed for routine diagnosis of bacterial, viral and fungal infections in various body fluids like Blood, CSF, Pleural, Sputum, Ascitic, Peritoneal, Synovial fluid etc. In recent years, chemically synthesized synthetic peptides have been shown to be popular tools for diagnosis of human infections. Peptide-based assay helps to differentiate subtle changes in monoclonal antibody specificities, and hence can be used as diagnostic biomarkers to provide a more differentiated serological diagnosis. Therefore, synthetic peptides are now widely used in diagnostic assays as compared to the whole antigen to detect antibodies directed against specific antigenic epitopes of the infecting organisms. Several immunodiagnostic tests based on synthetic peptides derived from carefully selected highly antigenic proteins have been devised for viral, bacterial diseases of humans in recent years. Synthetic Peptide-based assay helps to differentiate subtle changes in monoclonal antibody specificities and has the major advantage of being cost effective, than conventional antigen-based tests. Neurological disorders and diseases represent a significant cause of morbidity and mortality worldwide. Diagnostic tests are vital tools that help physicians confirm or rule out a neurological disorder /diseases along with severity which are difficult to rule out from radiological findings due to similar clinical manifestations especially in case of Bacterial / viral infections of CNS. Perhaps the most significant changes during the past 10-15 years have occurred in clinical diagnostics of CNS infections with number of immunological markers bursting into the scene that aid in early yet cost effective diagnosis for CNS diseases. In current workshop, we will include various immunological techniques used for diagnosis of Neurological infection and disorders at CIIMS. Through comprehensive technical events, workshop will provide detail and key insights to this technique and approaches used to design effective diagnostics for detection of CNS disorders and diseases. In addition we will conduct a special session on Statistical analysis for students to bolster scientific interest & their application in medical research.

Day 2

  1. Hands on training for Reagent Preparation, Micropipette handling, Multichannel pipette handling, Instrumentation handling
  2. Hands on training of Indirect ELISA (TB Multiplex antibodies ELISA)
  3. Comparison of means T-test, ROC curve, determination of sensitivity, specificity, cut-off, chi-square test, using MedCalc (version 10) statistical software
  4. Hands-on on Graphical representation of data using Graph pad prism
  5. Video lecture on Indirect ELISA
  6. Report preparation
  7. Presentation on Lateral Flow Immunoassays (LFA)
